Isolation & subculturing of early leaf spot, Passalora arachidicola, of peanut, by Maharjan Namrata
Early Leaf spot (ELS) is one of the important foliar diseases of peanut, caused by fungus Passalora arachidicola. This video demonstrates isolation and subculture of ELS fungus in V8 medium. First the video shows diagnosis and confirmation of the fungus based on morphology and its spores, followed by isolation and subculture. It also shows the general method of spore increment for large scale experiments. Large amounts of inoculum of spores are required to perform such experiments like a disease screening bioassay.
